  • 【摘要】The invention belongs to the field of cell biology, and relates to a method for separating and purifying Schwann cells by using collagenase and ferment enzyme and application thereof. The method comprises the following steps of: peeling an epineurium of a nerve fragment under the sterile condition, chopping, cleaning, centrifuging, and removing supernate then digesting nerve fragments for 30 to 60 minutes by using the ferment enzyme, the collagenase NB4 or mixed solution of the ferment enzyme and the collagenase NB4, performing repeated blowing, then centrifuging, and removing supernate to obtain a cell mass and finally, re-suspending the cell mass by using Schwann cell culture solution. In the method for separating and purifying the Schwann cells by using the collagenase and the ferment enzyme, after primary purification, the purity of the Schwann cells can reach 98 percent. After secondary purification and amplification, the purity of the Schwann cells can approach 100 percent. By using the method, a large amount of high-purity Schwann cells can be obtained with low cost and high efficiency and the method provides rich high-quality Schwann cells for injured nerve repair.
