【摘要】The invention belongs to the field of a micro-fluidic chip, and relates to an improved one-step method for preparing the micro-fluidic chip by a freeze drying process. The method comprises the step of fixing biomacromolecules on the surface of a chip mold at fixed points. According to the invention, in the fixed point fixing step, a spotting antibody is dried by the freeze drying process, then PDMS (Polydimethylsiloxane) is poured, after the PDMS pouring layer is solidified, demolding is carried out, and the fixed biomolecules are simultaneously removed. By adopting the freeze drying process, the antibody is completely, uniformly and efficiently fixed on the internal surface of the micro-fluidic chip and by adopting the method, the defects in the prior art are overcome, the high-quality micro-fluidic chip is simply, conveniently and rapidly prepared, the chip yield and detection sensitivity are improved and the cost is low.
